Position Summary

This scientific leadership role will be responsible for developing a new capability within Moderna for rational and computational molecular engineering, including co-optimization of primary sequence and higher level structure for both protein and mRNA. The goal is to understand and leverage the role of sequence and structure of both protein and mRNA in multiple relevant processes, spanning from mRNA production (purification, yield, formulation) through to in vivo therapeutic activity (stability, efficacy, immunogenicity, etc.). This will involve the use of established computational protein engineering techniques and the development of novel techniques for mRNA, and coupling them together to form a tightly integrated target optimization pipeline. This engine will be applied to multiple projects across Moderna, including partners, and venture companies.

Responsibilities
•Design, characterize, and optimize protein and mRNA for key programs, including monoclonal antibodies, antibody fragments, as well as non-antibody target proteins including various epitope tags, fusion proteins, etc.
•Develop and apply rational design techniques to both mRNA and protein to optimize key parameters
•Serve as a technical expert in molecular engineering within the organization and provide support to multiple projects
•Interact closely with internal groups in R&D and Production to advance projects investigating production efficiency, translation efficiency, immunological effects, etc.
•Identify, manage and contribute to external collaborations relevant to molecular engineering
•Maintain accountability for project success and results delivery
•Contribute to scientific and strategy discussions to advance and enhance the scientific platform and product candidates
•Communicate research and development findings internally and externally

Minimum Qualifications
•Ph.D. in computational chemistry, bioinformatics, computational biology, or related field
•5+ years of experience (including 3+ in industry) in the field of molecular engineering•Experience with antibody engineering required (humanization, CDR engineering, antigen epitope analysis, design of alternative antibody formats, etc.)
•Experience with nucleic acid macromolecules a strong positive
•Deep technical expertise required in at least one of (preferred in all of) computational chemistry, cheminformatics, and bioinformatics:Bioinformatics - sequence alignment, analysis, and manipulation, secondary structure prediction, and analysis of experimental data (e.g. antibody display)
•Computational chemistry - 3D structure visualization and analysis, comparative protein modeling, molecular mechanics calculations and simulations, conformational analysis, and docking simulations
•Cheminformatics: physiochemical property calculations, QSAR/QSPR, clustering
•Proficiency in at least one programming or scripting languages required (multiple preferred), e.g. Python, Java, C/C++, Perl, Bash, etc.
•Experience with commercial and open source software for computational chemistry, bioinformatics, etc.
•Strong foundation in at least one of chemistry, molecular biology, immunology, and enzymology required (multiple preferred)
•An established record of innovative research achievements in molecular engineering reported in top-tier journals and patent applications
•Strong written and oral communication skills, comfort with senior management interactions
•Positive team building and teamwork skills: Innate ability to influence, lead and inspire people, within function and beyond functional/company boundaries
