PROGRAM DESCRIPTION

The Neutrophil Monitoring Laboratory, Clinical Services Program (Applied/ Developmental Directorate) provides clinical and basic laboratory support to Laboratory of Host Defenses and Laboratory of Clinical Infectious Diseases, NIAID. Data generated in the NML is used in the diagnosis and characterization of patients with various immunodeficiences including chronic granulomatous disease (CGD), leukocyte adhesion deficiency (LAD), and hyper-immunoglobulinemia syndrome (JOB Syndrome).

JOB DESCRIPTION

The NML is a CLIA-certified laboratory and the work required of this position must be performed with a high degree of skill, accuracy, consistency, and attention to detail. The duties include: 1) isolation of plasma and leukocyte subpopulations from whole blood using different isolation protocols, 2) cryopreservation and archival of the isolated biological samples, 3) performance of various commercial ELISAs for the analysis of neutrophil granule proteins and acute phase proteins, 4) performance of complex CLIA-certified functional assays that include determination of O2- generation by cytochrome c reduction and nitroblue tetrazolium staining and determination of PMN adherence to extracellular matrices, 5) isolation of DNA from frozen leukocytes or from EBV-transformed B cell lines and the determination of DNA concentration and quality using either Nanodrop instrumentation and/or picogreen fluorescence analysis, 6) Stimulation of PBMCs with a diverse array of agonists of cytokine production and subsequent detection of pro-inflammatory cytokines using Meso Scale multiplex immunoassay plates, 7) maintenance of databases and inventories of samples to be stored in the Central Repository, and 8) operation, maintenance, and quality control of biosensitive equipment including: automated cell counters, spectrophotometer, cytometer, and plate readers. Work may involve the use of potentially hazardous chemical or biological materials or may be located in areas where such materials are used. Requires the use of personal protective equipment and adhere to strict safety protocols. Operates and maintains sensitive biomedical equipment. Exercises independent judgment within a defined work scope. Sets up and conducts experiments or production assays or tests using protocols specified by a program scientist. Compiles, analyzes, and reports scientific data to laboratory supervisors. Keeps exact and detailed records and documents laboratory operations and data collection. Maintains proper quality control checks for experiments or production assays or tests. Independently performs assay methods development and seeks advice when appropriate. Performs special duties as requested by laboratory supervisor. May include formal reporting of data including presentation and/or publications.

Qualifications

: BASIC QUALIFICATIONS

- Possession of a Bachelor degree from an accredited college/university in a field related to biomedical research or four (4) years related experience in lieu of a degree.
- Foreign degrees must be evaluated for U.S. equivalency.
- In addition to educational requirements, a minimum of two (2) years related biomedical experience.
- Candidate must possess an understanding of the principals involved in the preparation of reagents, including media and salt solutions, as well as adjusting cell concentrations for either cryopreservation or performance of functional assays.
- This position is subject to obtaining a Public Trust clearance.
