Job Description:
The Regeneron Genetics Center is a wholly-owned subsidiary of the Company organized to collaborate with health systems and research groups to elucidate, on a large scale, genetic factors that cause or influence a range of human diseases. Building upon Regeneron's strengths in mouse genetics and genetics-driven drug discovery and development, the Center will specialize in ultra-high-throughput exome sequencing and computational biology; discovery of genotype-phenotype associations through linkage to well-annotated de-identified patient electronic medical records; and validation of discoveries using Regeneron’s VelociGene® technology. Our interests encompass a breadth of different areas such as Mendelian and family frameworks, large-scale population genetics (both common and rare variants), and gene-gene interactions. Program goals include target discovery, indication discovery, and patient-disease stratification. Objectives include advancing basic science around the world through public sharing of discoveries, providing clinically-valuable insights to physicians and patients of collaborating health-care systems, and identifying novel targets for drug development.
The Head of Analytical Genetics will take on a senior leadership position in the Regeneron Genetics Center and hire and manage a team responsible for the statistical and bioinformatics analysis of human genetics studies. The Analytical Genetics team will lead the statistical design of studies and analysis of resulting genomic data starting with annotated VCFs through variant annotation and filtering to association analyses and identification of causal variants associated with disease. This team will support the broader goals of the department: to apply human genetics to target discovery, selection of indications for clinical programs, and patient/disease stratification through the use of genomic data. The position will require coordination and collaboration with other functional area heads within the department, research and clinical scientists at Regeneron, and external collaborators.
Additional Responsibilities:
• Hire and manage an analytical genetics team consisting of statistical geneticists, computational biologists, bioinformaticians, programmers, and other required staff.
• Primary responsibilities will be to lead variant interpretation, statistical genetics, and bioinformatics analysis.
• Such efforts will require close integration with the analysis pipelines team, which will be responsible for alignment and calling, and subsequent leadership of post processing variant interpretation including any genotype and phenotype association analyses.
• Work closely with Head of Translational Genetics to select projects and establish study designs, taking into account statistical genetics considerations of power calculations, population structure, genetic architecture/models of disease, and requirements for replication/validation.
• Lead genomic analysis efforts for projects driven through the genetics center budget, but also collaborate with colleagues in research and clinical for projects run through the center and requiring expertise and guidance on study design and data interpretation.
• Provide statistical genetics expertise for the department and consult on projects within other R&D departments at Regeneron including therapeutic focus areas, translational and predictive medicine, and clinical development.
• Survey the literature and conferences for discoveries and findings from the broader genetics community. Integrate such findings into study opportunities and considerations internally.
• Provide scientific management of collaboration projects.
• Communicate and report study findings and analyses with leadership, administration, and collaborators.
• Contribute to management of department goals, as well as team and individual development goals.
Requirements
Experience and Required Skills:
· PhD in relevant field, such as statistical genetics or bioinformatics, with at least 5-10 years' experience in human genetics research and a strong analytical genetics background required.
· Minimum of 3-5 years of experience providing strategic leadership to a mid-sized team.
· Involvement in or use of large scale WES/WGS projects and data sets such as GO-ESP, 1,000 Genomes Project, CHARGE consortium, Centers for Mendelian Genomics, or others.
· Expertise in statistical genetics, including proficiency in the relevant tools and methods. Experience with a breadth of study designs and analyses spanning family based studies, population based studies, unrelated case-control studies, imputation, and sophisticated genotype-phenotype association studies including burden tests and collapsing methods. Strong requirement for rare variant association analyses.
· Proficiency with various variant annotation and filtering tools for predicting deleterious or LOF mutations and ranking putative causal variants.
· Experience with multiple genetics platforms (chip-typing, WES & WGS, targeted sequencing, etc) as well as other genomic profiling technologies (microarrays, RNASeq, etc) and integrating such datasets to strengthen evidence for associations.
