Job Description:
The Staff Research Associate II (SRA II) position is part of a research program studying the molecular immunology and genetics of inherited disorders of the immune system, and how lymphocytes develop from blood-forming stem cells in the bone marrow. The individual will be a technician working in mouse and human molecular immunology and genetics. Our work involves maintaining and doing experiments with an immunodeficient mouse colony as well as purifying and culturing lymphocytes from human blood samples.

Our laboratory employs molecular cloning and gene expression with plasmids and lentiviruses, CRISPR/Cas9 gene editing, immune phenotyping by flow cytometry, and cell-based assays of lymphocyte function to define the mechanisms of primary immune disorders and better understand lymphocyte differentiation.

The SRA II will perform colony maintenance and experiments with inbred mice that are immune deficient. Some experiments will involve injecting human cells into highly immune compromised (such as NOD/scid/gc-) mice in which the human cells can mature into T and B cells. Experiments will include immunologic assays, sterile culture and differentiation of mouse and human hematopoietic cells and lymphocytes, flow cytometry, and analysis of results. Results must be clearly written up and kept in order. The individual will work with the supervisor to plan experiments and controls and will go over results with senior lab members and the supervisor. Day to day trouble-shooting for scientific and logistical laboratory matters. The SRA II will be assisting and teaching new lab members and trainees joining the research team. Updating and renewing mouse and human protocols will be required, as will liaison with the Lab Manager for ordering supplies and maintaining equipment. Excellent written and spoken English essential.

Required Qualifications:

• BA/BS degree with coursework in a related science and at least a year of recent, directly related work experience utilizing the techniques or methods required by the position; demonstrated technical expertise

• Experience with cellular and molecular studies, including using flow cytometry, western, northern, PCR, ELISA assays

• Technical skills in sterile tissue culture and experimental procedure

• Excellent written, verbal, organizational, presentation, and computer skills

• Ability to handle sensitive clinical information appropriately

• Ability to work with mice and with human & mouse specimens and cells

• Outstanding interpersonal skills and teamwork, time management to meet deadlines

• Willingness and ability to learn new skills for changing research priorities

Preferred Qualifications:

• M.S. degree in molecular biology, genetics or bioinformatics

• One or more years related experience in immunology, molecular biology, genetics or mouse research following degree completion

• Facility with genomic databases

• Ability to diagnose problems when something in the laboratory is not working; address and report as needed in a timely manner

• Ability to deal with collaborators and outside contacts

• Ability to perform basic statistical analyses

• Ability to search genomic databases, locate and describe DNA variants with correct terminology
