Prior experience and accomplishment with tissue culture, immunology and genetics protocols well as working with mice and with human samples is essential. Molecular cloning and gene expression with plasmids and lentiviruses, immune phenotyping by flow cytometry and functional testing are used to define the mechanisms of primary immune disorders. Appropriate controls must be included with each experiment. CRISPR/Cas9 gene editing will be performed on cell lines and primary cells. The SRA III will maintain a limited size colony and will perform experiments with inbred mice bearing specific immunodeficiency genes. Some experiments will involve injecting human hematopoietic cells into newborn highly immune compromised mice (such as NOD/scid/gc-) in which the human cells mature into T and B cells. Experiments will include immunologic assays, sterile culture and differentiation of mouse and human hematopoietic cells and lymphocytes, flow cytometry, and analysis of results. Results must be clearly written up and kept in order. The individual will work with the supervisor to plan medium and long range experiments as well as perform day to day trouble-shooting for scientific and logistical laboratory matters.
